Can someone please clearly explain the best way to perform a back-up of everything on Mantis? We have over 800 tickets in our system and it will be a disaster if we lost everything. I will relay the responses to our Network Admins.

If you store your attachments in database there is nothing more to do than to backup your database.
If you store your attachments on disk, you have to backup also your configured attachment director(y)(ies)
This is enough to store your data.
If you customized MantisBT by writing custom functions or plugins you should also backup this customization.
If not, a backup of file config_inc.php and writing a README with the version of MantisBT you use is enough.
To restore a certain version of MantisBT you can use http://sourceforge.net/projects/mantisb ... tis-stable but I would consider to backup the whole MantisBT installation (it's just about 12MB)
