When I try and create new global profiles I just get get a red "Access Denied" message. I am logged into the Admin account and using version 2.27.0.

You do need to activate Global profiles in your config:
$g_enable_profiles = ON;

Great! Any idea when that will come in?

There is no planned date for 2.27.1 at the moment.
You can manually apply the fix (just change one singe line) If it's urgent for you
