Hello,
We are running version 1.0.1 with a MySQL database and are planning to migrate to the latest 1.2.x version available. What is the best upgrade path for this? Should we upgrade via a series of steps to 1.2.15, for example? Or install 1.2.15 new? If we install 1.2.15 new, how can we handle migrating data from the old 1.0.1 system to the new 1.2.15 version?

Migration of the data is handled by the installation process

Thank you - can I upgrade directly from 1.0.1 to 1.2.x? If not, what intermediate steps would be needed?

I never tried myself, but upgrading should run in one step.
Additional steps are only needed for versions < 1.0.0.
It's documented in the admin guide, but let me tell it again:
Always Backup your code, data and config files before upgrading !
